Effects of Trifolium repens and Trifolium pratense on the LPS-induced Inflammatory Responses in RAW 264.7 Macrophages

Abstract

Inflammation is a defensive response to external stimuli such as wounds and infections. However, excessive inflammation causes chronic inflammatory disease through the overexpression of inflammatory mediators such as nitric oxide (NO) and prostaglandin (PG) E2, and inflammatory cytokines such as interleukins (ILs). This study aimed to investigate the anti-inflammatory effects of methanolic extract of clover (MEC) and red clover (MERC) on lipopolysaccharide (LPS)-stimulated immune responses in RAW 264.7 cells. MEC and MERC (0, 62.5, 125, 250, and 500 µg/mL) were pretreated for an hour before stimulation with LPS (1 µg/mL). MEC and MERC did not exhibit cytotoxicity under 500 µg/mL and inhibited the production of inflammatory mediators such as NO and PGE2. In addition, MEC and MERC suppressed the LPS-induced expression of proinflammatory cytokines, including IL-6 and IL-1β at the mRNA and protein levels in RAW 264.7. The overall findings of this study suggest that MEC and MERC can be used as treatments for various inflammatory diseases.
