Anti-obesity Effects of Treatment with Air-classified Brassica juncea L. Extract in High-Fat Diet-Induced Obese C57BL/6J Mice 고지방식이로 유도된 C57BL/6J 마우스에서 공기분급 갓분말 추출물의 항비만 효과

Abstract

Air-classified Brassica juncea L. extract (BJ) was investigated for its anti-obesity effects in C57BL/6J mice fed a high-fat diet (HFD). Mice were divided into two groups: one fed a normal diet (NFD), and the other, an HFD. Mice in the HFD-fed group were subsequently treated with BJ (50, 100, and 200 mg/kg/day) for 8 weeks. Inhibition of HFD-induced obesity by BJ treatment was evaluated by analyzing biochemical parameters and using real-time polymerase chain reaction assays. BJ supplementation significantly decreased the final body weight, adipose tissue size, and epididymal adipose tissue volume in comparison with those in the HFD-induced obese mice (p < 0.05). BJ treatment also decreased serum triglyceride, glucose, high-density lipoprotein cholesterol, and low-density lipoprotein cholesterol levels in obese mice. Furthermore, it attenuated HFD- induced obesity by downregulating C/EBPα, PPARγ, and SREBP-1c to suppress adipogenesis. Moreover, BJ treatment activated the AMPK pathway, increased ACC phosphorylation, and downstream CPT1 expression in HFD-induced obese mice. These results demonstrate that BJ has anti-obesity effects in vivo, indicating that it is an excellent candidate for developing anti-obesity functional foods.

본 연구에서는 공기분급 갓분말 추출물이 고지방식이 섭취 비만 마우스의 비만 및 지질대사 개선에 미치는 영향을 검토하였다. C57BL/6 마우스의 체중은 100 mg/kg/day부터 유의적인 체중 감소를 보였으며, 100와 200 mg/kg/day 투여에 의한 농도 의존적인 유의적 차이는 나타나지 않았다. 갓분말 추출물 100과 200 mg/kg/day 농도에서 간과 내장지방 무게, 혈중 중성지방 함량, 총콜레스테롤함량, LDL-콜레스테롤함량, 지방세포 크기가 효과적으로 감소되었으며, 특히, 갓분말 추출물 200 mg/kg/day 농도에서 HFD군과 비교했을 때 부고환지방무게가 현저히 감소함을 나타내었다. 이와 같은 결과는 공기분급 갓분말 추출물이 transcription factor인 PPARγ, C/EBPα와 SREBP-1c의 발현을 유의적으로 감소하는 것에 기인하는 것으로 판단되며, 또한 지방조직에서 adiponectin을 증가시켜 AMPK pathway를 활성화하여 지방조직의 에너지 대사 장애를 개선하는 것으로 사료된다. 한편, 본 소재와 관련된 기능성분 및 작용기작에 대하여 추후 보다 구체적인 연구가 요구되며 이를 통해 기능성과 경쟁력을 갖춘 간편식, 영양 균형식으로의 개발이 기대된다.
